Does the chalk only come off by brushing? Will it come off with soap and water?

It has always come off with just brushing for me. But, it may depend on how much you use and how fat your line, and how hard you press. On dark fabrics, I have wet my finger with a little water to remove it a little better. And, of course, it does wash away as well.
